James is still sporting a snarl when he sees Cooper approaching and speaks to him. James isn't sure what's going to happen but he's ready for a fight and that much is obvious from how vigorously he tossed the vampire Cooper encouraged to find greener pastures out.
"Am I going to have to do something else brave and stupid?"
It's his way of asking if Cooper is going to try anything - he can't tell good vampires from bad at this point and frankly he'd rather err on the side of paranoia than trust strangers at this point.
